Moore was educated at the Glasgow Grammar School and then the University of Glasgow where he studied medicine, literature, history and philosophy. He left Glasgow in 1747 and practised as a surgeon with the army in the Netherlands and as the surgeon to the Duke of Albemarle's household in Paris where he engaged in further study. He returned to Scotland in 1751 and became a member of the Faculty of Physicians and Surgeons of Glasgow the same year. He spent the next twenty years in a successful surgery practice in the city and graduated MD from the University of Glasgow in 1770.

Moore accompanied the 8th Duke of Hamilton on a Grand Tour of Europe from 1772 to 1778, and wrote two acclaimed volumes on European travel, novels, and later his impressions of events he witnessed during the French Revolution. He carried on a correspondence with the poet Robert Burns. He moved to London shortly after completing the Tour.

One of Moore's five sons was the celebrated General Sir John, the "Hero of Corunna".
